摘要 目的:研究人眼眶前脂肪细胞的培养方法,并比较不同方法培养的利弊。方法:分别采用组织块法和酶消化法对人眼眶前脂肪细胞进行原代培养。结果:两种方法均能培养出大量的前脂肪细胞,并能进行传代。组织块法方法简单,需要的组织标本量少,但在原代培养时组织块容易脱离瓶壁,生长欠均匀,传代前生长较缓慢,培养的细胞成分较混杂;酶消化法获得的细胞成分较单一,增殖迅速,但步骤较复杂,需要的标本量较多。结论:人眼眶前脂肪细胞可用组织块法和酶消化法进行原代培养,研究者可根据病例标本获得量的多寡选择不同的培养方法。 AIM: To study the methods in culturing human orbital preadipocytes, compare advantages and disadvantages of them. METHODS: Tissue culture methods and enzyme - digesting methods were used in the primary cell culture of human orbital preadipocytes. We compared these two methods through cells isolation, cells culture, immuno phenotypic analyses and adipogenic differentiation. RESULTS: These were two methods reliable to obtain enough preadipocytes which could proliferate and had limited passages. Tissue culture methods were simpler. But the sampies were easy to detach from the wall of the culturing bottles when culturing the primary cells. The cells grew slower and might not grow regularly in primary culture. Enzyme - digesting methods could obtain simpler cells than tissue culture methods and the primary cells proliferated rapidly, while the procedure was complicated and more samples were needed. CONCLUSION: Tissue culture methods and enzyme - digesting methods are both reliable for the primary cell culture of human preadipocytes. According to the quantity of the samples obtained, the researchers can choose different methods.
