丹参注射液促心肌梗塞后大鼠缺血心肌血管新生作用及对相关生长因子的影响研究 被引量:11

摘要 目的观察丹参注射液对心肌梗塞(MI)后大鼠缺血心肌血管新生作用,并探讨其机制。方法雄性Wistar大鼠,结扎左冠状动脉造成急性心肌梗塞(AMI)模型,随机分为丹参注射液组、麝香保心丸(阳性对照)组、模型组(阴性对照),并设假手术组,6周后处死动物。用免疫组织化学法,检测各组大鼠缺血心肌中微血管数(MVC)、微血管密度(MVD)及血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、血小板衍生生长因子β(PDGF-β)、胰岛素样生长因子(IGF-1)蛋白的表达,用病理图像分析系统测定生长因子蛋白表达灰度值并进行半定量分析。结果模型组、麝香保心丸组及丹参注射液组大鼠MI边缘区MVC和MCD较假手术组明显增多(P<0.05);丹参注射液组大鼠缺血心肌局部MVC和MVD明显高于模型组(P<0.05),与麝香保心丸组作用相似(P>0.05)。模型组VEGF,bFGF-β,IGF-1蛋白表达及其灰度值明显高于假手术组(P<0.05);丹参注射液组VEGF,bFGF,PDGF-β蛋白表达灰度值明显高于模型组(P<0.05),而IGF-1蛋白表达灰度值低于模型组(P>0.05)VEGF,bFGF,PDGF-β,IGF-1蛋白表达灰度值低于麝香保心丸组(P<0.05)。结论丹参注射液有促MI后大鼠缺血心肌血管新生的作用,其机制可能与促进VEGF,bFGF,PDGF-β的表达有关。 Objective To study the angiogenesic effects and mechanisms of Salvia miltiorrhiza Injection on the ischemic myocardium of the rats after myocardial infarction(MI). Methods Male wistar rats were ligated left coronary artery to be made into models of acute myocardial infarction (AMI). All rats were randomly divided into Salvia miltiorrhiza Injection, Shexiang baoxin pills (positive control group), model (negative contral group) group and sham operation group. Treated them respectively for six weeks. Microvascular count( MVC), microvascular density (MVD), vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), insulinkind growth factor (IGF) and platelet- derived endothelial growth factor (PDGF) in ischemic myocardium of MI rats were detected by immunobistochemistry assay. The values of protein expression of VEGF,bFGF, PDGF- β and IGF - 1 on ischemic myocardyzum were calculated and analied by image analysis system. Results There were more MVC and MVD in ischemic myocardium of AMI rats of model group,salvia miltiorrhiza injection group, Shexiang baoxin pills group than sham operation group. MVC and MVD were higher in ischemic myocardium of AMI rats of Salvia miltiorrhiza Injection group than those of model group. Compared with Shexiang baoxin pills, they had the same roll. The values of protein expression of VEGF, bFGF, PDGF- β and IGF - 1 in model group were significantly elevated compared with that in sham operation group. The protein expression of VEGF, bFGF and PDGF - β in salvia miltionrrhiza injection group was significantly elevated compared with that in sham operation group. The protein expression of VEGF,bFGF and PDGF - β in Salvia miltiorrhiza Injection group was significant- ly elevated compared with that in model group. IGF - 1 in Salvia miltiorrhiza Injection group was lower than modle group. But there was no significant statistics difference between them. There were significant statistics differences in the protein expression of VEGF, bFGF, PDGF - βand IGF - 1 in salvia miltiorrhiza injection group compared with that in Shexiang baoxin pills group. Con- clusion Salvia miltiorrhiza Injection has the effect of promoting angiogenesis of the ischemic myocardium in rats after MI. Its mechanisms can be related with stimulating the expression of VEGF,bGFG and PDGF- β.
