高渗刺激促进大鼠视上核酪氨酸蛋白的磷酸化

摘要 目的研究高渗刺激对大鼠视上核酪氨酸蛋白磷酸化的影响。方法成年SD大鼠随机分为高渗组和对照组(每组n=5),分别腹腔注射高渗盐水、生理盐水,2 h后用4%多聚甲醛灌注固定,取脑作30μm冰冻切片,进行磷酸酪氨酸和c-Fos蛋白的免疫组织化学染色。结果高渗组大鼠视上核神经元磷酸酪氨酸蛋白表达高于对照组(P<0.01)。结论高渗盐水刺激促进视上核神经元酪氨酸蛋白的磷酸化。 Objective To study the effect of hypertonic stimulation on the expression of phosphotyrosine in supraoptic nucleus of rat. Methods The adult SD rats were divided into a test group and a control group ( n = 5 each) to receive intraperitoneal injection of hypertonic saline and normal saline respectively. The rats were infused with 4% paraformalde- hyde 2 h after the injection, and then, the brains were removed to prepare 30 pan frozen sections. The expression of phosphotyrosine, as well as that of c - Fos, in the supraoptic nucleus was detected by immunohistochemistry. Results The expression of phosphotyrosine was higher in the hypertonic saline test group than in the control (P 〈0.01 ). Conclusion Hypertonic stimulation enhances the expression of phosphotyrosine in supraoptic nucleus of rat.
