核苷二磷酸激酶等蛋白在人胰腺癌与癌旁组织中的差异表达 被引量:1

摘要 目的应用比较蛋白质组学方法研究人胰腺导管腺癌组织和癌旁组织之间差异表达的蛋白质。方法利用双向凝胶电泳(2-DE)对8例胰腺导管腺癌组织和癌旁组织的总蛋白进行分离,两组间差异蛋白质点用质谱和数据库检索鉴定。利用蛋白质印迹分析和免疫组织化学法检测差异蛋白在胰腺癌和癌旁组织的表达。结果鉴定出30个差异表达蛋白,包括亲环素A、过氧化氧化还原蛋白1、14-3-3sigma、AnnexinII、热休克蛋白27、核苷二磷酸激酶(NDPK)等。差异表达蛋白NDPK在35例胰腺癌组织表达率为77.1%,而配对癌旁组织为28.6%。结论2-DE为基础的蛋白质组学技术是研究肿瘤的一种重要手段。NDPK等差异表达蛋白可能是潜在的胰腺癌诊断标记物。 Objective A comparative proteomic approach was used to identify and analyze proteins related to pancreatic cancer. Methods Proteomes of eight pairs of pancreatic ductal adenocarcinoma tissue samples and normal adjacent tissue samples were obtained by two-dimensional gel electrophoresis (2-DE). Comprehensive analyses of 2-DE proteins patterns between the two groups were done using computerized image analysis. Selected proteins exhibiting statistically significant alternations were identified by mass spectrometry. In addition, Western blotting and immunohistochemistry (IHC) were performed to verify the expression of certain candidate protein in pancreatic cancer tissue and normal adjacent tissue. Results A total of 30 protein spots, including cyclophilin A, peroxiredoxin 1, 14-3-3 sigma, annexin II, heat shock 27 000 protein, nucleoside diphosphate kinase (NDPK), were identified. The expression rate of NDPK in 35 samples of pancreatic cancer tissues was 77. 1%, compared with 28.6% in normal adjacent tissues. Conclusions The analysis of proteomics with 2-DE in human tissue is useful for discovering potential cancer marker candidates. These differentially expressed proteins (NDPK) may be potential biomarkers for early detection of pancreatic cancer.
