摘要 目的探讨缺氧后乳鼠心肌细胞中热休克蛋白90a(hsp90a)表达及变化规律,探讨hsp90a在缺氧心肌细胞中发挥的保护作用。方法采用建立的心肌细胞缺氧模型,蛋白免疫印迹法测定hsp90a蛋白表达变化水平,免疫组化法显示心肌细胞中hsp90a表达的定位。结果缺氧后乳鼠心肌细胞中hsp90a的表达明显升高且呈现出一定规律性。结论缺氧可以诱导心肌细胞中hsp90a的表达增加,其主要分布于心肌细胞浆内,作为一个重要的调控分子参与早期缺氧心肌细胞的保护机制。 Objective To investigate the expression and change of heat shock protein 90a (hsp90a) in the neonatal rats' cardiac cells after ischemia, and discuss the protective effects of hsp90a in the ischemic cardiac cells. Methods Using the ischemic cardiac cells' model founded by our experimental rooms, we measured the changing level of hsp90a expression by western-blotting, and displayed the sites of hsp90a expression in the cardiac cells by immunohistochemistry. Results hsp90a expression was obviously upregulated in neonatal rats' cardiac cells after ischemia, and hsp90a was found mainly in the cytoplasm. Conclusions Ischemia could induce high expression of hsp90a in the cardiac cells, and hso90a was involved in the protective mechanism of the early ischemic cardiac cells as an important regulating factor.
