摘要
目的分析探讨短棒状杆菌(Cp)化学成分及抗骨髓瘤的抑瘤效果。方法用Lowry法测定Cp的蛋白质含量;用蒽酮法测量Cp的多糖含量;用6-氨基喹啉基-N-羟基琥珀酰亚胺基-氨基甲酸酯(AQC)柱前衍生荧光法对Cp水解液中各种游离氨基酸种类及含量分析;小鼠腹腔注射骨髓瘤细胞,观察Cp抑制骨髓瘤及延长生存期的效果。结果Cp中蛋白质含量为562.08±18.26μg/mg;多糖含量为174.77±2.63μg/mg;除色氨酸被水解而未被检出外,Cp水解液中含有17种氨基酸,其中有7种人体必需氨基酸;对骨髓瘤有明显的抑瘤效果。结论Cp制剂中含有丰富的蛋白质、多糖成分,且具有很好的抑制骨髓瘤的效果。
Objective To investigate the chemical components of Corynebacterium parvum (Cp) and its inhibit effect on the growth of myeloma in mice. Methods Protein content of Cp was determined with Lowry's method; polysaccharide content was determined with anthrone method; free amino acids in Cp hydrolysate were analysed with 6 -aminoquinolyl -N -hydroxysuccinimicly carbamate( AQC ) precolumn fluorescene derivatization. Myeloma ceils were injected into the abdominal cavity of the mice and the inhibit effect of Cp on the growth of myeloma was observed, Results Protein content in Cp was 562, 08 ± 18.26 μg/mg and polysaccharide content was 174, 77 ± 2, 63 μg/mg; There were 17 kinds of amino acids (including 7 kinds of essential amino acids) in Cp, except that tryptophan was destroyed by hydrolysis, The growth of myeloma ceils were significantly inhibited by Cp. Conclusion Cp has high protein and polysaccharide content and can significantly inhibit the growth of myeloma in mice.
