紫外线作用肿瘤细胞后基因表达谱的变化

The Change of Gene Expression Profile in Cancer Cells Receiving Ultraviolet Irradiation

目的分析紫外线(UV)对人原发性肝癌细胞株基因表达谱的影响。方法以限制片段差异显示聚合酶链反应(RFDD-PCR)技术分析紫外线照射人原发性肝癌细胞株前后基因表达谱的变化。结果获得表达序列标签共257条。其中差异表达序列标签116条,含开启表达序列标签38条,关闭表达序列标签23条,表达增强序列标签40条,表达降低序列标签15条。结论紫外线可引起人原发性肝癌细胞株基因表达谱发生变化,这些表达发生变化的基因在肿瘤生物学中的作用有待进一步研究。

Objective To analyze the effects of ultraviolet irradiation on gene expression profile of human primary hepatic carcinoma cell line. Methods The variation of gene expression profiles between cell lines with and without ultraviolet radiation was observed by carrying out Restriction Fragment Differential Display-PCR （RFDD-PCR）. Results A total of 257 expressed sequence tags （EST） were obtained, in which we got 116 differential expression EST, including 38 open EST, 23 close EST, 40 increase EST and 15 decrease EST. Conclusion Ultraviolet may change the cell line gene expression profile of human primary hepatic carcinoma, the roles of these differential expression genes will be studied in future.
