摘要
目的研究冷冻治疗对大鼠C6脑胶质瘤的增殖抑制作用及其机制。方法建立大鼠C6脑胶质瘤模型,60只载瘤大鼠随机均分为对照组和治疗组。治疗组实施冷冻治疗后免疫组化检测P27Kip1蛋白的表达,磁共振检测肿瘤体积变化,并观察动物生存期。对照组未实施冷冻治疗。结果与对照组比较,冷冻治疗可上调P27Kip1蛋白的表达,对照组为(4.81±0.72)%,治疗组为(28.84±3.56)%,P<0.05;缩小肿瘤体积,对照组为(292.97±19.13)mm3,治疗组为(76.37±10.22)mm3,P<0.05;延长动物生存期,对照组为(37.00±1.55)d,治疗组为(57.00±3.16)d,P<0.05。P27Kip1蛋白阳性表达率和荷瘤鼠生存期呈正相关(r=0.836,P<0.05);同体积呈负相关(r=-0.696,P<0.05)。结论冷冻治疗可抑制大鼠C6脑胶质瘤的增殖,延长动物生存期,其机制同上调P27Kip1蛋白的表达有关。
Objective To investigate the effect of cryotherapy on C6 rat brain glioma and the mechanism. Methods C6 rats' intracerebral glioma models (60 rats) were established and equally randomized to control group and treatment group. After cryotherapy, the expression of P27Kipl protein was defected by immunohistochemical staining, the tumor size was measured by MRI and the rat survival time was observed in the treatment group. The control group did not receive cryotherapy. Results Compared with the control group, cryotherapy up-regulated the expression of P27Kipl protein (The control group 4.81±0.72% vs treatment group 28.84±3.56%) (P〈0.05), and reduced the tumor size (control group 292.97±19.13mm^3 vs treatment group 76.37±10.22mm^3) (P〈0.05), and prolonged the rat survival time (control group 37.00±1.55 days vs treatment group 57.00±3.16 days) (P〈0.05). A positive correlation existed between the expression of P27Kipl protein and life span (r=0.836, P〈0.05),while the expression of P27Kipl protein and tumor size showed a negative correlation (r=-0.696, P〈0.05). Conclusion Cryotherapy can inhibit the growth of C6 rat brain glioma and prolong survival time of the rat, and the mechanism is concerned with the up-regulation of P27Kipl protein.
