摘要
目的 探讨如何建立适合长期治疗的胶质瘤动物模型.方法 将C6胶质瘤株移植到雌性Balb/c无胸腺小鼠右肩皮下建立6只荷瘤鼠模型,观测肿瘤生长曲线、苏木素和伊红染色病理组织切片、流式细胞仪碘化丙啶染色液染色细胞周期划分及凋亡分析、免疫组织化学法测定胶质纤维酸性蛋白.结果 6只雌性Balb/c无胸腺小鼠右肩皮下均生长出持续增大的肿瘤(100%),生长曲线符合三次方程曲线(P<0.001).苏木素和伊红染色显示符合恶性星型细胞瘤的表现.流式细胞仪碘化丙啶染色液染色显示大部分肿瘤细胞处于G0~G1期(90.26±3.15)%,S期(7.71±1.11)%,G2-M期(2.03±0.79)%.免疫组织化学法测定胶质纤维酸性蛋白,显示胞浆强阳性,证实为神经胶质来源.结论 皮下移植肿瘤细胞于裸小鼠形成的胶质瘤动物模型可靠且稳定.
Objective To make an animal model of malign glioma for lorig-term treatment. Methods C6 malign glioma cells were inoculated into endermic tissues of 6 Balb/c female nude mice, and the tumor growth was observed. HE staining, propidium iodide (PI) staining flow cytometry analysis and immunohistochemical staining were used to study the expresions of p53 and glial fibrillary acid protein(GFAP). Results All the 6 Balb/c female nude mice showed tumor growth without atrophy(100%). The growth curve was in accord with cubic equation(P〈0, 001). By HE staining,we found that the tumor tissue was malign glioma,most cells were GFAP positive. The tumor cells were most in G0-G1 period (90. 26± 3.15%),and there were(7. 71±1.11)% in S period and(2. 03±0.79)% in G2-M period. Conclusion Planting C6 malign glioma cells to nude mice endermic tissue can make a stable and feasible animal model of malign glioma.
