摘要
首先从分类地位、毒粒结构、基因组结构、基因组转录调节及复制繁殖周期等方面对人类腺病毒分子生物学进展作了概述,然后论述了人类腺病毒用作基因工程载体的特性。人类腺病毒不仅是用作研究真核基因表达调控机制的良好模型,而且是基因工程的重要载体。人类腺病毒载体除具有组建多价重组口服活疫苗的前景外,还可以作为一种表达载体获得高水平的外源蛋白,研究组织特异性表达和建立稳定的细胞系,以及在机体组织中转移表达报告基因以治疗疾病等。
After shortly reviewing the moleculor biological development of human adenovirus from the following respects of viral taxonomy,viral structure,genomic structure,genomic trans- criptional regulation and viral productive replication,this paper has comprehensively discussed the specific property of human adenovirus as genetic engineering vectors.Human adenoviruses are not only good models for mechanism investigation of real nucleus gene expression and its modulation, but also important genetic engineering vectors.Human adenovirus vectors have the prospects of constructing multivalent recombinant live-vaccines which can be orally used.As expression vec- tors,recombinant human adenovirus vectors can also be used for expressing high levels of foreign proteins,studying tissue-specific gene expression,establishing stable cell lines for gene expres- sion,transfering and expressing reportor genes in body tissues in viro to cure some diseases.
