摘要
目的:探讨明胶白芨胶/纳米血竭多孔材料对大鼠创面组织血管内皮细胞生长因子(VEGF)表达的影响。方法:选用沉淀法制备血竭纳米混悬液,应用冷冻干燥法制备明胶白芨胶/纳米血竭多孔材料,制作大鼠背部全层皮肤缺损动物模型,分为材料治疗组、空白对照组和材料基质对照组,分别用明胶/白芨胶纳米血竭多孔材料、无菌纱布(用注射用水浸湿)、明胶/白芨胶多孔材料(不含纳米血竭)覆盖创面,于致伤后7d取材,免疫组织化学染色(SABC法)检测明胶/白芨胶载药多孔材料对创面组织VEGF的表达。结果:明胶/白芨胶血竭多孔材料组大鼠皮肤VEGF免疫组化染色细胞平均吸光度值较无菌纱布组及明胶/白芨胶多孔材料组高(p<0.01,p<0.05)。结论:明胶/白芨胶血竭多孔材料能促进创面肉芽组织及毛细血管生长,能促进创面组织VEGF的高表达。
Objectives:To explore the Effect of gelatin-Bletilla hyacinthine gum carrying nano Sanguis Draconis on expression of VEGF in rat wound. Methods: The precipitation method was used to prepare nano Sanguis Draconis suspl, and it was compounded with scaffolds (gelatin and BHG). Then it was frozen and dried to form spongy and porous material. The full skin loss of rats were prepared, the wounds were covered by Porous Biomaterial carrying TCM(gelatin-Bletilla hyacinthine gum carrying nano Sanguis Draconis), sterility bandage, Porous Biomaterial (gelatin-Bletilla hyacinthine gum) respectively. The rats were sacrificed at 7d after the injury. The expression of VEGF in the wound tissue were detected by immunohistochemical techniques. Results:The increase of VEGF was observed at 7d after injury at the group of gelatin-- Bletilla hyacinthine gum carrying nano Sanguis Draconis, Compared with sterility bandage group and gelatin-Bletilla hyac inthine gum group, it showed obvious difference. (p〈0.01,p〈0.05) Conclusion: The biomaterial carrying nano Sanguis Draconis prepared in this study can promote the growth of micrangium by the higher level VEGF expression.
