内皮素-1在大鼠肢体缺血再灌注后肝损伤中的变化及其意义 被引量:2

摘要 ①目的探讨内皮素-1(ET-1)在大鼠肢体缺血再灌注后肝损伤中的变化及其意义。②方法实验用雄性Wistar大鼠,随机分为对照组、肢体缺血再灌组(LIR组)和BQ-123(内皮素受体特异性阻断剂)+LIR组,每组8只。分别测定各组动物血浆ET-1、透明质酸(HA)、丙二醛(MDA)、谷丙转氨酶(ALT)、谷草转氨酶(AST)及肝组织ET-1、MDA、髓过氧化物酶(MPO)的含量和DNA双链百分率的变化。③结果LIR组与对照组比较,血浆ET-1、HA、MDA、ALT、AST和肝组织中ET-1、MDA、MPO含量升高,肝组织DNA双链百分率下降(P<0.05),组织损伤加重。BQ-123+LIR组与LIR组比较,血浆ET-1、HA、MDA、ALT、AST和肝组织中ET-1、MDA、MPO含量下降,肝组织DNA双链百分率升高(P<0.05),组织损伤减轻。④结论大鼠LIR后继发性肝损伤可能与肝微循环障碍有关。 Objective To evaluate the possible role of endothelin - 1 ( ET - 1 ) in the liver injury after hind limbs ischemia - reperfusion in rats. Methods Wistar rats were divided into 3 groups ( per group n = 8) : control group, limbs ischemia - reperfusion group (LIR) and BQ - 123 ( ET receptor antagonist) + LIR group. The changes of ET - 1, hyaluronic acid ( HA ), malondialdehyde (MDA),alanine transaminase(ALT) and aspartate transaminase(AST) in plasma were observed. The contents of ET- 1, MDA, Myeloperoxide (MPO) and the ratio of DNA chain in liver were detected. Results Compared with the control group, the levels of ET - 1, HA, MDA, ALT and AST in plasma and the contents of ET - 1, MDA, MPO in liver increased, and the ratio of DNA chain in liver decreased in the LIR group( P 〈 0.05 ). When BQ - 123 group was compared with LIR group, the levels of ET - 1, HA,MDA,ALT and AST in plasma and the contents of ET- 1, MDA and MPO in liver decreased,while the ratio of DNA chain in liver increased( P 〈 0.05 ) , and liver injury lightened in BQ - 123 group. Conclusion The microcirculation disturbance in liver might be the cause of liver injury after hind limbs ischemia - reperfusion in rats, and ET - 1 mould participate in it.
