摘要
目的探讨乳腺癌接种后裸鼠后血清中基质金属蛋白酶2的含量变化,为诊断乳腺癌早期转移提供新的依据。方法将乳腺癌细胞接种裸鼠成瘤后,观察乳腺癌成瘤过程,并采用酶联免疫吸附试验(ELISA)检测肿瘤形成过程中裸鼠血清中MMP-2含量的变化,并结合乳腺癌生物学行为进行分析。结果乳腺癌MCF-7细胞在接种后第9天至第11天后,于接种部位出现肉眼可见肿瘤结节,随时间推移进行性生长,血清中MMP-2含量明显增高,血清中MMP-2的含量与肿瘤浸润深度、肿瘤组织Ⅳ型胶原分布、淋巴结转移关系密切。结论乳腺癌动物模型血清中MMP-2含量变化与肿瘤浸润关系密切。测定血清中MMP-2含量可作为判断乳腺癌早期转移的临床观察指标之一。
Objective To establish a breast carcinoma model in nude mice to investigate the matrix metalloproteinase-2 (MMP- 2) content change in serm during the invasion and metastasis of cancer cells, and to analyze the relationship between MMP-2 content. Methods Breast carcinoma nude mice was established to study the tumor generation and growth ; ELISA method was used to test 1MMP-2 content in serm of breast carcinoma model in nude mice during the different points of time; clinical data analysis was used to research on the relationship between MMP-2 and cancer biological character. Results In the group of 1 x 106/ml breast cancer cells, a single tumor clot which can be seen by naked eyes appeared at 9 - 11 days after inoculation. With the time going on, the volume of the clot became larger and larger. The MMP-2 content in serm extremely increased continuously. The MMP-2 content was closely related to the depth of invasion, Col Ⅳ morphous and lymph nodes status of cancer cells. Conclusion The whole proceed of cancer matastasis is accompanied by the increase of MMP-2 proteolytic activity and the degradation of extracellular matrix. This model is feasible to study the metastasis character of breast cancer. The MMP-2 content in breast cancer nude mice' s sera has significant correlation with the depth of invasion, tissue differentiation and lymph nodes status of the cancer cells.
