摘要
选取犊牛大网膜脂肪组织,进行牛前脂肪细胞的单层贴壁培养,在细胞接种后第4、6、8、10、12、141、6 d分别提取细胞总RNA,采用实时荧光定量RT-PCR方法,检测了体外培养的犊牛前脂肪细胞不同时期ADPN基因mRNA和HSL基因mRNA的表达情况。结果表明,单层培养的脂肪细胞在第4、6、8、10、12、141、6 d,ADPN基因mRNA的表达水平呈先下降后升高的趋势,而HSL基因mRNA的表达水平则呈下降趋势。
Preadipocytes from epiploon of newborn calf were cultured by monolayer adherent method, and total RNA of the preadipocytes was extracted on day 4,6,8,10, 12,14 and 16 post-culture, respectively. The abundance of ADPN mRNA and HSL mRNA was detected by real-time fluorescent quantitative RT-PCR during differentiation period from preadipocyte to adipocyte. The results showed that the abundance of ADPN mRNA was up-regulated during the differentiation period(on day 2 to 12), but was down-regulated during post-differentiation (on day 14 to 16), and HSL mRNA was decreased on day 10- 16 post-cultivation.
