摘要
用化学方法合成了硝基无色孔雀石绿(NO2-LMG),还原为氨基无色孔雀石绿(NH2-LMG),经红外光谱扫描鉴定NH2-LMG为所需化合物。氨基无色孔雀石绿经重氮法处理后与兔血清白蛋白(RSA)载体蛋白相连,通过紫外分光光谱鉴定,偶联产物与其前体物相比吸收峰发生了飘移,说明偶联成功。通过动物免疫试验和ELISA抗体特异性试验对该人工合成抗原免疫原性和与无色孔雀石绿反应原性的鉴定,表明成功合成了无色孔雀石绿免疫原。
NO2-LMG was synthesized by chemical method and reduced to NH2-LMG. Then, NH2-LMG was identified by the infrared spectrometry(IRS). NH2-LMG treated with diazotization was linked with rabbit serum albumin(RSA),which was proved using the UV spectrophotometer scanning absorption peak floating. The immunogenicity and reactogenicity for the LMG of artificial antigen were examined using animal immunity test and ELISA test. The result showed that the LMG-RSA immunogen was successfully obtained.
