β-细辛醚对谷氨酸所致PC12细胞损伤的保护作用 被引量:12

摘要 目的研究β-细辛醚对谷氨酸所诱导的PC12细胞损伤的保护作用,并探讨其作用机理。方法用谷氨酸造成PC12细胞损伤模型,观察β-细辛醚对其细胞形态、损伤程度、存活力、钙离子浓度和细胞凋亡的影响。结果10mmol/L谷氨酸作用PC12细胞16h,出现明显损伤,凋亡率和死亡率升高,培养上清液中LDH含量增加;7.5、15、30μg/mLβ-细辛醚可有效改善谷氨酸损伤引起的PC12细胞形态改变,提高细胞存活力,减少乳酸脱氢酶渗漏;15、30μg/mLβ-细辛醚能明显降低谷氨酸引起的PC12细胞内钙离子浓度和细胞凋亡率。结论β-细辛醚对谷氨酸所致PC12细胞损伤具有保护作用,其作用机制可能与钙拮抗作用有关。 Objective To study the protective effects of β-asarone on PC12 cells damage induced by Glutamate. Method The effects of β -asarone on PC12 cells after Glutamate intoxication on morphology, extent of damage, livability, intracellular calcium concentration and apoptosis ratio were observed. Result Morphological changes, LDH leakage and intracellular calcium concentration increasing, and cell survival decreasing were observed in PC 12 cells exposured to Glutamate. 7.5, 15, 30 μg/mL β-asarone can increase cell survival, decrease LDH leakage. 15, 30 μg/mL β-asarone can reduce intracellular calcium concentration and apoptosis ratio. Conclusion β-asarone prevents the toxicity of Glutamate, and it maybe attribute to its effect of anticalcium.
