摘要
目的研究哇巴因对乳鼠心室肌起搏电流电生理特性的作用。方法体外培养乳鼠心室肌细胞。一周之内,应用常规全细胞膜片钳技术检测1,20,40,80μmol/L哇巴因对培养心室肌细胞起搏电流的电流密度、起搏阈值等的作用。结果1μmol/L的哇巴因即可明显增加起搏电流的电流密度(4.76±0.48pA/pFvs4.0±0.41pA/pF,P<0.01)。随着哇巴因浓度进一步增加,起搏电流的电流密度逐渐增大,HCN通道的半最大激活电位逐渐向去极化方向改变,电导曲线明显右移,活化速度也明显加快,尾电流被增强,但反转电位不受影响。结论哇巴因对心室肌HCN通道可能有剂量依赖性的激活作用。
Objective To observe the effect of ouabain on pacemaker currents in isolated cadiomyocytes of SD rat. Methods The single ventricular myocytes of neonatal SD rats were cultured . The whole-cell patch clamp recording technique was used to record the change of pacemaker current by different dosages of ouabain from 1 to 80 μmo//L. Results Ouabain iucreased current density significantly by a dose-dependent manner, even at concentration of 1 μmol/L(4.76 ± 0.48 pA/pF vs 4.0± 0.41 pA/pF, P 〈 0.01 ). The V0.5 altered to depolarization direction, current conductance curves shifted to right and active velocity increased significantly with the action of ouabain. The pacemaker currents almost recovered to original level after the washout of ouabain. Ouabain increased amplitude of tail current, but had no influence on reversal potential.
