摘要
目的应用整体可视化成像技术观察Tiam-1基因沉默对人大肠癌细胞株SW480转移特性的影响。方法将绿色增强荧光蛋白(EGFP)标记的人大肠癌细胞株SW480/EGFP+与SW480/EGFP+/Tiam-1-经裸鼠尾静脉注射和结肠原位接种,分别建立可视化转移动物模型,比较两种细胞株成瘤与转移特性的差别。结果SW480/EGFP+与SW480/EGFP+/Tiam-1-细胞均稳定表达EGFP;SW480/EGFP+/Tiam-1-的Tiam1基因干扰率达70%;SW480/EGFP+/Tiam-1-和SW480/EGFP+比较体外增殖能力无显著差异(P>0.05),但体内增殖能力差异显著(P<0.05);SW480/EGFP+/Tiam-1-转移率明显降低。结论Tiam1基因可能在人大肠癌侵袭转移中发挥重要作用。
Objective To observe the effect of Tiam-1 gene silencing on the metastasis of human colorectal carcinoma cell line SW480 in nude mice by real-time whole-body fluorescence imaging. Methods Enhanced green fluorescence protein (EGFP) -labeled human colorectal carcinoma cells, SW480/EGFP^+/Triam-1^- and SW480/EGFP^+, were implanted into nude mice via tail vein injection or orthotopic colonal inoculation, and real-time whole-body fluorescence imaging was performed to compare the difference in tumor progression and metastasis between the two cells. Results Both SW480/EGFP^+ and SW480/ EGFP^+/Tiam-1^- cells stably expressed EGFP, and Tiaml gene expression was reduced in SW480/EGFP^+/Tiam-1^-to 30% of the expression level in SW480/EGFP^+ cells. The growth rate of the two cell lines had no significant difference in vitro (P〉0.05), but SW480/EGFP^+/Tiam1^- cell proliferation and metastasis were depressed obviously in comparison with SW480/EGFP^+ in vivo (P〈0.05). Conclusion Tiam-1 gene may play an important role in invasion and metastasis of human colorectal cancer.
