摘要 目的研究AG-490拮抗谷氨酸对PC12细胞中P-STAT3活性的上调作用及其抗凋亡意义。方法谷氨酸诱导PC12细胞凋亡。Hochest荧光染色观察PC12细胞形态。流式细胞仪(FCM)分析评价凋亡百分率。Westernblot测定P-STAT3蛋白表达量,MTT检测PC12细胞存活率。结果谷氨酸诱导PC12细胞中STAT3蛋白表达上调(P=0.000),如予Jak/STAT3信号通路阻断剂AG-490预处理,则其表达被下调(P=0.002),细胞凋亡率减低(P=0.000),细胞存活率升高(P=0.000)。结论AG-490拮抗谷氨酸诱导的PC12细胞凋亡,其机制可能与STAT3活化有关。 Objective To investigate the rivalry effects of AG - 490 with glutamic acid on the activation of P - STAT3 which resists apoptosis of PCl2 cells. Methods Glutamic acidwas used to inctuce apoptosis of PC21 cells. Cell morphology was observed by Hochest fluorescent staining. Apoptosis percentage was analyzed by flow cytometry. P - STAT3 protein expression was detected by Western blotting, and survival rate of PC12 cells was detected by M-I-F. Results Protein expression of STAT3 up- regulated in PC21 cells induced by glutamic acid(P= 0. 000). AG - 490 was used to block Jak/STAT3 signal access, thus the protein expression clown- regulated, apoptosis rate of PC12 cell reduced and survival rate improved. Conclusion AG- 490 can antagonize the glutamic acid induced apoptosis of PCl2 cells, and the underlying mechanism might be concerned with activation of STAT3 protein.
