摘要
以干制五倍子为原料,采用L16(44)正交试验法,详细分析了鞣质溶剂浸提的主要影响因素,并通过溶剂萃取、Ca2+离子沉淀、明胶沉淀等不同分离纯化方法对鞣质最终得率和纯度进行了比较研究,初步建立了五倍子鞣质的提取工艺。实验结果表明:以100%丙酮为提取溶剂、温度50℃、pH2.0、时间10min为五倍子鞣质的最佳提取条件,采用乙酸乙酯萃取的鞣质最终得率较高,且鞣质纯度达到了89.15%。
The optimum extraction conditions of tannin from galla chinensis were studied by using orthogonal test L16(4^4). Then ethyl acetate extraction, gelatine precipitation and Ca^2+ precipitation studied separation and purification of Tannin. The optimum extraction process conditions were as follows. 100% acetone as extracting agent, extracting temperature 50℃, pH value 2.0, required time 10min. The results show that acetone as extraction agent and the ethyl acetate extraction, the purification degree was up to 89.15%. The yield was also relatively high ethyl acetate extraction.
