摘要
膦甲酸钠毒性试验结果表明:小鼠经静脉注射LD50为395.7mg/kg;大鼠经腹腔注射LD50为1050.5mg/kg;大鼠腹腔注射、狗静脉注射3个月长期毒性试验,无毒反应剂量分别为150mg/kg、60mg/kg,毒性反应剂量分别为300mg/kg、120mg/kg,主要中毒表现为肝、肾功能异常,中毒性肾病及肾小管坏死等;Ames试验中膦甲酸钠对鼠伤寒沙门菌无致突变作用;微核试验中膦甲酸钠未诱发NIH小鼠微核率的增加。
Trisodium Phosphonoformate (Foscarnet) is a new antiviral drug. The results of acute toxicity study showed that the LD50 volue of Fascarnet caused by intravenous injection(iv) was 395. 7mg/kg for mice, and 1050. 5mg/kg for rats by intraperitoneal injection (ip). Ninety day feeding test was found that the non-toxic reaction doses were 150. 60mg/kg and the toxic reaction doses were 300.120mg/kg for rats by ip and dogs by iv,respectively. The cardinal toxic symptoms were hepatic and renal dysfuction, toxic nephropathy,renal tubules necrosis et al. At the same the,we also found Foscarnet had no mutagenic action in Ames test,and did not increase the micronuclei frequence in micronucleus test with NIH mouse.
