五味子对去卵巢小鼠的记忆保持和海马NOS神经元表达的影响 被引量:7

摘要 目的:研究五味子(Fructus Schisandrae Chinensis)改善记忆与类雌激素作用的关系,并探讨相关的作用机制。方法:采用一次性被动回避反应行为学方法研究五味子对去卵巢小鼠记忆能力的影响;NADPH-d酶组化染色观察五味子对小鼠海马亚区一氧化氮合酶(NOS)阳性神经元形态及数目的影响。结果:去卵巢可明显降低小鼠的记忆能力,同时降低海马各区NOS阳性神经元数目。五味子醇提物具有改善去卵巢小鼠记忆保持的能力,显著增加海马各区NOS阳性神经元数目;五味子脂提物却明显抑制去卵巢小鼠的记忆保持,相应降低海马各区NOS阳性神经元的数目。结论:五味子改善记忆保持与类雌激素作用有关,其类雌激素作用的主要成分存在于乙醇提取物中,但五味子脂提物存在拮抗类雌激素的作用。影响海马各区NOS阳性神经元的合成及分布可能是五味子醇提物的类雌激素作用机制之一。 Objective: To investigate whether the memory improvment of Fructus Schisandrae Chinensis is contribute to estrogen-like action and the underlying mechanism. Methods: Step-through test and NADPH-d histochemical staining were used to evaluate the memory ability and observe the number of NOS positive neurons in the hippocampus. Results: The memory of ovariectomized mice was impaired significantly and the number of NOS positive neurons was decreased in the subregions of hippocampus compared to that of control group. The ethanol extracts of Fructus Schisandrae Chinensis enhanced the memory of ovariectomized mice and increased the number of NOS positive neurons in hippocampus significantly. However, petroleum ether extracts of Fructus Schisandrae Chinensis significantly decreased the memory of ovariectomized mice and further decreased the number of NOS positive neurons in hippocampus. Conclusion: The memory improvement of Fructus Schisandrae Chinensis is related to estrogenlike action, and the effective components exists in ethanol extracts. However, some anti-effective components might be concentrated in the petroleum ether extracts. The estrogen-like action of ethanol extracts of Fructus Schisandrae Chinensis might affect the synthesis and distribution of NOS positive neurons in various subregion of hippocampus.
