低强度脉冲超声作用下兔髁突软骨生长改建的半定量组织学研究

摘要 目的:评价低强度脉冲超声(LIPUS)对兔髁突软骨生长改建的影响,探讨LIPUS作为一种功能性矫形治疗辅助手段促进髁突生长改建的可行性。方法:选择6只10周龄的新西兰雄性大白兔,左侧髁突,低强度脉冲超声局部辐照,20min/d×4周,右侧作为实验对照侧。处死后取双侧髁突行HE染色,并采用JD801形态学图像分析系统,获得髁突软骨各层层厚及总层厚、单位面积细胞数目及单个细胞面积的半定量测量结果。结果:①.实验侧髁突软骨的总层厚为467.31um,与对照侧总层厚368.38um相比,二者具有显著性差异;②.厚度变化主要集中在生发层、过渡层及成熟层,尤其以成熟层厚度变化最为明显。③.与对照侧相比,实验侧生发层、过渡层及成熟层中单位面积细胞数目均有显著性增加,尤其以生发层和过渡层最为明显;④.与对照侧相比,实验侧过渡层、成熟层及移行层中单个细胞面积均有显著性增加,其中以成熟层的变化最为明显。结论:LIPUS能够有效刺激兔髁突软骨的生长改建,作为一种功能性矫形治疗辅助手段促进髁突生长改建具有一定的可行性。 Objective: The purpose of this study is to evaluate the modification of rabbit condylar cartilage under the use of low-intensity pulsed ultrasound.Method: Six growing New Zealand malerabbits (10 weeks of age)were chosen for this study.LIPUS was applied for 20 minutes per day for four weeks to the left side of the mandible in each rabbit, while the right side served as thecontrol side. Then 4-urn-thick sections were cut along the sagittal plane, and stained with hematoxylin and eosin. JD801 Morphology Software was used to measure the changes. Students' t-test was used to determine if there were significant differences between the 2 sides.Result: (1).There was significant difference between the experimental side (ES)and control side (CS) in the total layer thickness:467.31 um to 368.38 urn. (2).The most obvious change of layer thickness was in maturation layer. (3).There were significant differences between ES and CS in the cell amount of unpolarization, transition, and maturation layers, especially of the former 2 layers. (4) .There were significant differences between ES and CS in the cell area of transition, maturation, and calcification layers, especially of the maturation layer.Conclusion:LIPUS can effectively stimulate the modification of rabbit condylar cartilage,which indicates the feasibility of LIPUS as an assistant supplement of traditional functional therapy.
