摘要
[目的]研究β-榄香烯对肝星状细胞(HSC)凋亡及凋亡相关蛋白的影响。[方法]传代培养的大鼠HSC系HSC-T6与不同剂量β-榄香烯(终浓度为10、5、2.5 mg/L)共同培养48 h后,应用Annexin-V-碘化丙锭染色检测β-榄香烯对HSC凋亡的影响,免疫组化检测凋亡相关蛋白BcL-2/Bax。[结果]β-榄香烯可使HSC凋亡率明显增多,同时可使凋亡抑制分子Bcl-2表达下调,促凋亡分子Bax表达增强(P<0.01)。[结论]β-榄香烯可通过下调Bcl-2/Bax比率,促进HSC凋亡。
[Objective]To investigate the effect of β-elemene on apoptosis and Bcl-2/Bax expression of the hepatic stellate cells(HSC) in vitro. [Methods]The rat hepatic stellate cells line- HSC-T6 was incubated with various concentrations of beta-elemene for 48 hours. The apoptosis was examined with Annexin V-Propidium iodide assay, and the expression of Bax, Bcl-2 was detected with cell immunocytochemistry stain. [-Results] β-elemene could markedly induce the apoptosis of the cells with Bcl-2 protein expression down-regulated and Bax protein expression up-regulated(P〈0. 01). [Conclusion ]β-elemene could down regulate the ratio of Bcl/Bax to increase the sensitivity of apoptosis and induced HSC apoptosis.
