老龄心肌梗死大鼠肝叶切除围手术期血小板GPⅡb/Ⅲa受体的变化

Changes of Platelet Glycoprotein Ⅱb/Ⅲa Receptors During Perioperative Period of Hepatolobectomy in Aged Rats With Myocardial Infarction

摘要 目的:观察老龄心肌梗死大鼠肝叶切除围手术期血小板表面GPⅡb/Ⅲa受体的动态变化。方法:老龄大鼠26只,随机分为2组,对照组及心肌梗死组各13只。异丙基肾上腺素连续3d腹腔内注射,制作老龄大鼠心肌梗死模型。3周后行肝叶切除术,以PE标记的CD61荧光抗体流式细胞技术检测两组术前、后72h内血小板表面GPⅡb/Ⅲa受体的变化。结果:老龄心肌梗死大鼠肝叶切除术后6、12、24、48及72h血小板表面GPⅡb/Ⅲa受体密度均较术前明显升高(P均<0.05)。而对照组仅于术后12h较术前升高。结论:肝叶切除术可导致老龄心肌梗死大鼠血小板表面GPⅡb/Ⅲa受体密度增高,是围手术期血栓前状态形成的原因之一。 Objective: To observe dynamic changes of platelet glycoprotein Ⅱ b/Ⅲ a receptors during perioperative period of hepatolobectomy in aged rats with myocardial infarction. Methods: Twenty six aged SD rats were randomly divided into two groups, control group and infarction group, with 13 rats in each group. Aged models with myocardial infarction were induced by intraperitoneal injection of isoproterenol for consecutive 3 days. All rats underwent hepatolobectomy. Changes of GP Ⅱ b/Ⅲ a receptors on platelet surface were detected by flow cytometric analysis before and 6, 12, 24, 48 and 72 hours after operation. Results: Density of GP Ⅱ b/Ⅲ a receptors increased continuously in myocardial infarction group 6, 12, 24, 48 and 72 hours after operation compared with preoperative period (P〈0. 05). The density of GPⅡ b/Ⅲ a receptors increased only at 12 hours after operation in control group compared with that of preoperative period (P〈0. 05). Conclusions: Hepatolobectomy lead to an increase density of platelet GP Ⅱ b/Ⅲ a receptor in aged rats with myocardial infarction during the perioperative period and is one of the reasons for perioperative pre-thrombotic state.
