摘要
目的:探讨过量维甲酸(RA)对金黄地鼠胚胎神经管维甲酸受体α(RARα)基因和蛋白表达的影响。方法:采用原位杂交和免疫组织化学方法及图像分析,半定量分析金黄地鼠胚胎神经管正常发生及RA致神经管畸形过程中RARα转录和表达水平的变化。结果:正常对照组RARα表达水平随胚龄增加而逐渐下降。实验组在灌服过量RA后3 h,RARα表达水平明显低于正常对照组;6 h时其表达水平明显高于正常对照组;14、22、46 h其表达水平下降,与对照组相比无显著差异;96 h其表达增强,并显著高于对照组水平。结论:过量RA引起的RARα转录和表达水平的变化与RA致金黄地鼠神经管畸形有关。
Objective: To investigate the effect of excess retinoic acid (RA) on the expressions of RAreceptor a (RARa) in neural tube of golden hamsters. Methods: Using in situ hybridization, immunohistochemistry and image analysis methods, the difference of RARa expressions between the normal and defected neural tubes induced by excess RA was analyzed in golden hamsters. Results: The expression of RARa in the normal embryonic neural tube tissues was time dependant. After being fed with retinoic acid, the expression level of RARa was significantly lower at 3 h and higher at 6 h than that of the corresponding control group; then the expression levels decreased at 11, 22, 46 h, without significant differences compared with the corresponding control group. Till to 96 h, the expression increased again and was much higher than that of the corresponding control group. Conclusion: Changes in RARa expression induced by excess RA may be related to neural tube defects in golden hamster.
