摘要
目的:研究p27^(kip1)和Skp2在正常和坐骨神经夹伤后大鼠脊髓中的表达变化。方法:Westem印迹法,免疫组织化学,免疫荧光双标。结果:Western印迹结果显示坐骨神经夹伤后,脊髓中p27^(kip1)表达下降后又有所恢复,并且p27^(kip1)和Skp2在脊髓中的表达变化呈负相关。免疫组织化学及免疫荧光双标显示,p27^(kip1)和Skp2在正常和损伤后的脊髓神经元和胶质细胞中都有表达。坐骨神经夹伤后p27^(kip1)和Skp2在脊髓腹角阳性细胞数目变化呈负相关。结论:坐骨神经损伤后p27^(kip1)。和Skp2在脊髓腹角神经元及其周围的胶质细胞表达变化相关,对研究脊髓损伤和修复的分子机制有重要意义。
Objective: To investigate expressions of p27kip1 and Skp2 in normal and injured rat spinal cords. Methods: Western blot,immunohistochemistry and double irnmunolabelling were used. Results: Expressions of p27kip1 and Skp2 were detected in rat spinal cords. The level of p27kip1 in the spinal cord was declined after sciatic nerve crush, and then it was rescued. And the changes of p27kip1 and Skp2 in rat spinal cords after sciatic nerve injury were inversely correlated. It was showed that p27kip1 and Skp2 were expressed in the neurons and gliocytes. Statistical analysis indicated that there was negative correlation between the numbers of p27kip1 and Skp2 positive cells in the ventral horn after sciatic nerve injury. Conclusion: After sciatic nerve injury, the expressions of p27kip1 and Skp2 were correlated tightly in the neurons and the glial cells around in the ventral horn of the spinal cord, which is of great significance for the investigation of the molecular mechanism of renovation of the spinal cord after peripheral nerve injury.
