制川乌配伍白芍的增效作用及其作用机理 被引量:26

摘要 目的从镇痛、抗炎、免疫调节3个方面研究制川乌配伍白芍的增效作用及其机理。方法采用扭体、热板、痹证足部压痛、耳廓肿胀、痹证足部肿胀、痹证免疫脏器试验;中医风寒湿痹证模型检测炎性因子(PGE2)、一氧化氮(NO)、类风湿因子及细胞因子(IL-1β、TNF-α、IL-6)、单胺类递质(5-羟色胺、多巴胺、去甲肾上腺素、5-羟吲哚乙酸)等指标来揭示作用机理。结果制川乌配伍白芍能升高小鼠化学刺激、热刺激疼痛和大鼠痹证足部疼痛模型痛阈值,抑制小鼠二甲苯所致的耳廓肿胀和痹证大鼠足部肿胀,纠正痹证大鼠免疫脏器的病理变化;能升高痹证大鼠血浆中PGE2、血清中NO、血清细胞因子和脑组织单胺类递质的含量,降低痹证大鼠血清类风湿因子的含量。结论制川乌配伍白芍可部分增加制川乌镇痛、抗炎、免疫调节等作用。其抗炎机理可能与血浆中PGE2、血清中NO的释放有关;免疫调节机理可能是抑制类风湿因子和增加细胞因子来加强免疫作用;镇痛机理是可能增加中枢镇痛物质的合成。 OBJECTIVE To study synergistic action of the compatibility of Radix Aconiti preparata and Radix paeoniae alba on relieving pain, anti -inflammatory and immune-regulation, and reveal the mechanism. METHODS The stretching body test, the hot plate test, the feet tenderness test of arthromyodynia rats, the test of swelling of auricle, the feet swelling test of arthromyodynia rats, and the immunization organ test of arthromyodynia rats were used for the investigation. Arthralgia model according to traditional Chinese medical theory of wind - cold - wetness was established, to reveal the effect mechanism by detecting a series of indexes, e.g. inflammatory factor ( PGE2 ) , free radical(NO) , rheumatoid factor ( IgG), cell factor ( IL - 1 β, TNFα, IL - 6) and monoamine transmitter (5 - HT, DA, NE,5 - HIAA). RESULTS The compatibility of Radix Aconiti preparata and Radix paeoniae alba could increase the pain threshold value of the chemical stimulus, the thermal stimulus and the feet of arthromyodynia rats, restrain the swelling of mice auricle and the feet swelling of arthromyodynia rats. It could increase the content of PGE2 , NO, cell factor( IL - 1β, TNF-α, IL -6) in blood serum and brain tissue monoamine transmitter(5 -HT, DA, NE,5 -HIAA)of arthromyodynia rats, and decline the content of blood serum IgG. CONCLUSION Synergistic action of relieving pain, anti - inflammatory and immune - regulation can be partially proved by the compatibility of Radix Aconiti preparata and Radix paeoniae alba. The mechanism of anti - inflammatory may be related to the releasing of PGE2 and NO in blood serum. The mechanism of immune - regulation may be due to restraining of rheumatoid factor and increasing of cell factor. The increased synthesise of the central algogenic substance may be the mechanism of relieving pain.
