摘要
目的了解肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)启动子上游-1031位点基因多态性在中国汉族正常人群中的分布特点。方法应用聚合酶链反应(PCR)和限制性片段长度多态性(RFLP)方法对正常个体基因组DNA中TNF-α-1031位点基因多态性进行了观察,探讨其基因型及等位基因分布特点。结果正常汉族人群中TNF-α-1031位点基因型以TT型发生频率最高(64.4%),TC型较少(33.0%),CC型最少(2.6%),T等位基因的频率为80.9%,C等位基因的频率为19.1%,这种分布特点在正常男女间差异不显著(P〉0.05)。与其它种族相比较,TNF-α-1031位点基因型在中国正常人群中的分布与日本人群中的分布相似(P〉0.05)。结论中国汉族正常人群中存在TNF-α-1031多态性,其分布特点与日本人群中的分布相似。
Objective To investigate the polymorphism at -1031 position in the promoter region of the tumor necrosis factor-α (TNF-α) in HuBei population. Methods We used PCR- RELP to examine the distribution of TNF- α -1031 polymorphism in HuBei Han population and compared with Japanese. Rusults The TNF- α -1031 allelic frequencies were 80.9% for T allele and 19.1% for C allele in HuBei Han population. The genotypic frequencies were 64.4% for homozygous T/T allele, 33.0% for heterozygous for T/C allele and 2.6% for homozygous C/C allele. The TNF-α-1031 allelic frequencies in Chinese were the same as in Japanese. Conclusion The TNF- α-1031 polymorphism exists and was the same as in Japanese.
