芪丹泡腾片对大鼠脑缺血再灌注损伤的保护作用

摘要 目的观察芪丹泡腾片(QDPTP)对大鼠脑缺血再灌注损伤的保护作用。方法观测QDPTP对局灶性脑缺血再灌注模型大鼠的神经行为评分、脑梗死范围和脑含水量变化的影响;对全脑缺血再灌注损伤模型大鼠脑组织SOD活力、MDA含量以及脑组织和血清中IL-1β水平的影响。结果QDPTP能明显改善局灶性脑缺血再灌注引起的大鼠神经行为评级,显著降低脑梗死范围和脑含水量;亦能拮抗全脑缺血再灌注引起的SOD活力下降及MDA含量升高,高剂量(900mg/kg)组还能使脑组织和血清中IL-1β水平显著下降。结论QDPTP对大鼠脑缺血再灌注损伤具有保护作用,其机制可能与抗自由基损伤和抑制炎性细胞因子有关。 Objective To investigate the protective effect of QDPTP on cerebral ischemia reperfusion injury in rats. Methods The model of rat focal cerebral injury was established by middle cerebral artery occlusion-reperfusion to evaluate the behavior, infarct size and water content in brain tissue. The model of rat global cerebral ischemia reperfusion was made by means of Pulsinelli's method, and the SOD activity and MDA content in brain tissue as well as the IL-1β content in cerebrum and blood serum were measured. Results In focal cerebral injury rats, QDPTP (low and high dose) could significantly improve the behavior (all P〈0.05) and markedly decrease infarct size (all P〈0.01) as well as water content (P〈0.01 or P〈0.05). The drug could also enhance the SOD activity (all P〈0.01) and reduce the MDA content (all P〈0.01) in brain tissue. At the same time, the high dose of QDPTP could obviously lower the IL-1β content (P〈0.01) in cerebrum and blood serum. Conclusion QDPTP can reduce the free radical injury and inhibit the production of proinflammatory cytokine, which was responsible for the protective effect on cerebral ischemia reperfusion injury in rats.
