摘要
目的 采用HPLC法测定广西粉葛中葛根素的含量。方法 色谱柱Phenom enex Gem in i C18柱(250 mm×4.6 mm,5μm);流动相:乙腈(A)-0.4%磷酸水溶液(B),采用梯度洗脱;流速:1.0 m l/m in;检测波长:250 nm;柱温:30℃。结果 葛根素在800-2400 ng范围内峰面积与进样量呈良好的线性关系。回归方程为A=7.682×10^4C+5.885×10^4,r=0.999 3(n=5)。其平均回收率为99.49%,RSD为0.75%(n=5)。结论 该方法简便易行,结果准确,可作为广西粉葛药材质量控制的有效方法。
Objective To determine the content of puerarin from Guangxi Radix puerariae thomsonii by HPLC. Methods The isolation was conducted on a phenomenex gemini C18 column (250 rmm × 4.6 mm , 5 μm ) , the mobile phase consisted of acetonitnle (A) -0.4% phosphoric and (B) in a gradient model, the flow rate wes 1.0 ml/min, the detection wavelength was 250 nm and the column temporature was 30℃. Results The content of puerarin showed good linearity in the range of 800 ng -2400 ng,the regression equation was A =7. 682 × 10^4C +5. 885 × 10^4 ,r =0.999 3 (n =5), and the average recovery was 99.49%, RSD was 0.75% (n = 5 ). Conclusion This method is simple, rapid, accurate and reliable. It can be used for quality control of Guangxi Radix puerariae thomsonii.
