摘要
在单因素实验和Box-Behnken实验设计基础上,采用响应面分析法,确定胭脂虫中红色素的最优提取条件。验证实验显示,得到的最优条件能够很好符合预测值。通过响应面实验及回归分析,得到了胭脂虫红色素最佳工艺参数:温度40℃,乙醇体积分数60%,提取3次,每次提取时间0.54 h,每次提取液料比(体积∶质量)为(2∶1)mL/g。在该条件下提取率可达30.62%,提取回收率可达99.80%。取得的最佳工艺条件可以很好地用于胭脂红色素的生产,具有一定的理论和实用价值。
Based on the single factor experiments and the Box-Behnken design, response surface methodology was used to determine the optimal extraction condition. The validation test indicated that the optimal condition could ideally match the prediction. Through response surface methodology and regression analysis, the desired conditions were concluded : 40 ℃, alcohol volume fraction 60%, total extracting time 0.54 h, thrice for extraction, ratio of alcohol to material (2:1 ) mL/g. The present results show that the extraction rate can reach 30.62%, recovery rate can be 99.80%. The optimal parameters can be well used in the manufacture of cochineal pigments which has a certain theoretical and practical value.
