摘要
目的:本实验旨在研究Rh2对肿瘤细胞生长的抑制作用及其机理,以进一步阐述Rh2在抗肿瘤方面的作用。方法:Rh2作用于MCF-7及MCF-7/ADM细胞,应用MTT法确定各组细胞增殖抑制率。流式细胞仪分析Rh2导致MCF-7和MCF-7/ADM细胞坏死及凋亡的作用,及其对细胞周期的影响,并检测Rh2对MCF-7和MCF-7/ADM细胞内Caspase3活性的影响。结果:Rh2可以显著抑制MCF-7和MCF-7/ADM的增殖(P<0.05),并有时间和剂量依赖关系,对MCF-7抑制作用更强。Rh2可以导致MCF-7坏死,通过Caspase途径诱导MCF-7/ADM凋亡(P<0.05),对两种细胞周期的影响也有所不同。结论:人参皂甙Rh2通过不同机制抑制了MCF-7和MCF-7/ADM细胞的增殖,Rh2促进耐药细胞系MCF-7/ADM凋亡的研究结果显示其在抗肿瘤作用方面的独特性。
Objective To research on the mechanism of inhibitory effect of ginsenoside Rh2 on human mammaxy adenocareinoma MCF-7 and the drug resistance cell line MCF-7/ADM in vitro Methods In MTF tests, Rh2 were added to MCF-7 and MCF-7/ADM, inhibition ratios were determined. In flow cytometric analysis, necrotic and apoptotic effect of Rh2 on MCF-7 and MCF-7/ADM were determined. Effects of Rh2 on cell cycle were determined too. The activity of caspase 3 in MCF-7 and MCF-7/ADM cell lines was also detected. Results Ginsenoside Rh2 could inhibit the pro- lifemtion of MCF-7 and MCF-7/ADM cells with time and dose-dependent( P 〈 0.05) ,Rh2 had stronger effect on MCF-7 cells. Rh2 could cause necrosis on MCF-7, and induce apoptosis on MCF-7/ADM with Caspase pathway. The effects of Rh2 on Cell cycles on MCF-7 and MCF-7/ADM were different too. Condusion Ginsenoside Rh2 can inhibit the pro- lifemtion of MCF-7 and MCF-7/ADM cells with different mechanism. Rh2 showed individuality on anti-tumor effects.
