即时荧光定量PCR检测吉西他滨诱导胰腺癌Panc-1细胞凋亡通路关键基因ASK1的高表达

摘要 目的 检测抗癌药吉西他滨(gemcitabine)诱导胰腺癌Panc-1细胞凋亡的关键基因ASK1的高表达。方法 应用即时荧光定量PCR技术检测50.33μg/mL吉西他滨作用于胰腺癌Panc-1细胞48h后凋亡基因ASK1表达的变化。结果 吉西他滨作用于胰腺癌Panc-1细胞48h后凋亡基因ASK1高表达,比值为2.64E-02/1.25E-02=2.112,与基因芯片结果2.107622018非常接近。结论 吉西他滨诱导胰腺癌Panc-1细胞凋亡,ASK1高表达。 Objective To detect key apoptosis-related gene ASK1 of gemcitabine-mediated apoptosis on human pancreatic carcinoma Panc-1 cells. Methods The key apoptosis-related gene ASK1 on pancreatic carcinoma Panc-1 cells exposed to gemcitabine (50.33μg/mL) for 48h was detected by real-time fluorescent PCR. Results In contrast, these treated cells overexpressed ASK1 gene, its ratio was 2.64E-02/1.25E-02= 2.112. The value was the same as gene chip value(2.107622018). Conclusions Gemcitabine might induce the Panc-1 cell appoptosis and increase the expression of ASK1 gene.
