摘要
目的通过观察电针对脑缺血再灌注大鼠缺血区皮质TNF-α mRNA的表达和TNF-α含量的影响以探讨电针对缺血性脑损伤的保护机制。方法采用大脑中动脉线栓法制作模型;电针大鼠的水沟、内关,运用逆转录-多聚酶链反应(RT-PCR)法检测缺血区皮质TNF-α mRNA表达和放射免疫法检测皮质TNF-α含量。结果针刺各组缺血皮质中TNF-α mRNA表达和TNF-α含量与模型组相比均降低,有统计学意义。结论电针通过抑制TNF-α mRNA的表达减少TNF-α合成,以对抗缺血再灌注脑组织神经损伤。
Objective: To observe the effect of electroacupuncture(EA) on the expression of TNF -α mRNA and the level of TNF -α in ischemic penubra in cortices of rats with ischemia - reperfusion, so that probe the mechanism of EA against cerebral ischemia injury. Methods: EA on rats' Shuigou(GV26) and Neiguan(P6). Measured the level of expression of TNF -α mRNA by RT- PCR, the level of TNF -α in cortices by radioimmunoassy(RIA). Results: The expression of TNF -α mRNA and the level of TNF -α in cortex of EA groups were lower, comparing with those of the model groups. Conclusion: EA can lighten the neurons injury after cerebral ischemia- repeffusion by decreasing the expression of TNF -α mRNA in the ischemia region and the creation of TNF -α in cortex.
