摘要
目的观察黄芪对大鼠脑缺血再灌注后神经细胞凋亡及其相关基因JUN蛋白表达的影响,探讨其对脑缺血再灌注损伤保护作用的机制。方法分别采用TUNEL及免疫组织化学与医学图像分析相结合的方法检测大鼠脑组织神经细胞凋亡及JUN蛋白的表达。结果与假手术组相比,模型组大鼠脑组织神经细胞凋亡数目及JUN蛋白表达增多;与模型组相比,黄芪组大鼠脑组织神经细胞凋亡数目及JUN蛋白表达减少。结论黄芪可通过抑制脑缺血再灌注后神经细胞凋亡而发挥脑保护作用,其抗凋亡机制可能与下调JUN蛋白的表达有关。
Objective: To investigate the effiects of astragus on neuronal apoptosis and the expression of JUN protein after cerebral ischemia and reperfusion, probe the mechanism of astragus Preventing against cerebral ischemia- reprefusion injury. Methods: Methods of TUNEL and immunohistochenfistry were used respectively to measure neuronal apoptotmc cells and the expression of JUN protein in rats'brains. Results: Compared with those in the sham - operated group ,the numbers of neuronal apoptotic and JUN protein positive cells increased markedly in the model group. Compared with those in the model group, the nunlbers of neuronal apoptotic and JUN protein positive cells were less in the astragus group. Conclusion: Astragus could decrease neuronal apoptosis after cerebral ischemia and reperfusion by inhibiting the expression of JUN protein.
