摘要
目的探讨肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-9(MMP-9)和基质金属蛋白酶组织抑制因子-1(TIMP-1)在寻常型银屑病发病机制中的作用及其相关性。方法采用免疫组织化学SABC法检测40例寻常性银屑病皮损组织、20例非皮损组织以及20例正常皮肤组织中TNF-α、MMP-9和TIMP-1的表达分布情况。结果TNF-α、MMP-9在银屑病皮损组织中阳性表达率明显高于非皮损组织和正常皮肤组织(P<0.05);TIMP-1在皮损组织中的阳性表达率与非皮损组织和正常皮肤组织中的表达差异无显著性(P>0.05)。在银屑病皮损中,TNF-α与MMP-9表达呈正相关(r=0.471,P<0.01);MMP-9与TIMP-1表达呈负相关(r=-0.589,P<0.01);TNF-α与TIMP-1表达呈负相关(r=-0.6,P<0.01)。结论TNF-α、MMP-9和TIMP-1可能在寻常型银屑病发病机制中起相互协同作用。
Objective To study the role of TNF-α , MMP-9 and TIMP-1 in the lesions of psoriasis vulgaris. Methods SABC immunohistochemical technique was used to examine the expression of TNF-α, MMP-9 and TIMP-1 in lesional skin of 40 cases and uninvolved skin of 20 cases of psoriasis and in skin of 20 cases normal controls. Results The expression of TNF-α and MMP-9 was significantly higher in the lesional skin than that in nonlesional skin and normal controls (P〈0. 05). TIMP-1-positive rate in the involved psoriatic skin was not significantly higher than that in uninvolved and normal skin (P〉0.05). There was a positive correlation between the expression of TNF-α and MMP-9 ( r = 0.471, P 〈0.01 ), a negative correlation between the expression of MMP- 9 and TIMP- 1 ( r = -- 0.589, P〈0. 01), and also a negative correlation between the expression of TNF-α and TIMP-1 ( r = --0.6, P〈0. 01) in the skin lesions. Conclusion Overexpression of TNF- α , MMP-9 and TIMP-1 in psoriatic lesions may contribute to the pathogenesis of psoriasis.
