摘要 目的:研究膀胱癌相关Livin基因在膀胱移行细胞癌组织中的表达以及Livin基因表达与膀胱移行癌生物学行为的关系;研究抑癌基因p53在膀胱癌组织中突变以及Livin表达与p53基因突变的关系。方法:100例膀胱移行细胞癌组织,设计Livinα引物,以半定量RT-PCR的方法,检测膀胱癌组织Livinα mRNA,以免疫组化法检测癌组织中p53蛋白。将膀胱肿瘤病理分级、临床分期、是否复发等生物学行为及p53蛋白染色强度分别量化,与组织Livinα mRNART-PCR产物量进行等级相关分析。并以10例正常膀胱黏膜作为对照。结果:100例膀胱移行细胞癌,67例检测到Livinα mRNA表达,10例正常膀胱黏膜组织未检测到Livinα mRNA表达,Livinα mRNA在膀胱移行细胞癌组织中表达率高于正常膀胱黏膜,χ2=14.4,P=0.000。Livinα mRNA在膀胱癌组织中表达与p53蛋白阳性(rs=0.465,P=0.002)及肿瘤病理分级(rs=0.463,P=0.002)有相关性。结论:膀胱移行细胞癌组织中可见Livinα mRNA高表达,Livinα mRNA表达与膀胱癌病理分级及癌组织中p53蛋白呈正相关。 OBJECTIVE: To investigate Livin gene expression in bladder transitional cell carcinoma and explore its relationship with p53 gene expression in bladder cancer and clnicopathological futures of these lesions. METHODS: Reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemical staining were employed respectively to evaluate the Livin a mRNA expression and the p53 protein expression in 100 samples of bladder cancer, together with 10 samples of normal bladder tissue. RESULTS: Of the 100 bladder cancers, Livin a mRNA expression was detected in 67 cases, whereas, no Livin a mRNA expression was found in 10 normal bladder tissue. The positive rate of Livin a mRNA expression in bladder cancer was statistically higher than that in normal bladder tissue, x^2= 14.4, P= 0. 000. The Livin α mRNA expression was correlated with p53 protein expression (rs= 0. 465, P= 0. 002) and the bladder cancer pathological grade (rs= 0. 463,P=0. 002). CONCLUSION: The data of this study suggests that Livin α mRNA expression may be an objective parameter to predict the biological behavior and prognosis of bladder cancer.
