摘要
通过PCR方法从甘蓝型油菜基因组DNA中克隆出△12-脂肪酸脱氢酶基因BnFAD2的cDNA序列,全长1155bp,编码384个氨基酸,测序结果表明该基因与GenBank已登录的甘蓝型油菜FAD2基因(AF243045)的相似性为99%。将其克隆到植物表达载体pZP212上,利用农杆菌介导法转化烟草NC89品种,经卡那霉素筛选和PCR鉴定,得到阳性植株15棵。
Full- length of △^12 -fatty acid desaturase FAD2 cDNA with 1155 nucleotides was cloned from Brassica napus DNA by PCR, coding for 384 amino acids. Sequence alignment showed that the cloned fragment shared 99% of homology with Brassica napus (AF243045) from GenBank. The BnFAD2 was cloned into pZP212 vector. The plant expression vector was transformed into Tobacco NC89 mediated by Agrobacterium tumefaciens. 19 kanamycin - resistant transgenic tobaccos were obtained and fifteen of them were identified as positive lines by PCR.
