摘要
目的研究乙烯硫脲(ETU)致神经管畸形胎鼠大脑皮质中nNOS和iNOS表达情况,进一步探讨nNOS和iNOS在脑发育中的作用。方法应用免疫荧光、Western-blot及Real-time PCR方法检测nNOS和iNOS在正常胎鼠、ETU致畸的神经管畸形胎鼠的大脑皮质表达情况并进行定位、定量分析。结果nNOS在给药无畸形组、脊柱裂畸形组胎鼠大脑皮质中的表达与正常对照组相比无明显差异(p>0.05),而在脑膜膨出组胎鼠大脑皮质中nNOS的表达与正常对照组明显减少(p<0.01);iNOS在各组均未检测到。结论nNOS在脑膨出的胎鼠大脑皮质中表达明显减少,提示nNOS参与了脑异常发育过程。
Objective To explore the nNOS and iNOS expressions in the cerebral cortex of animal model with neural tube defects(NTDs). Method The immune-fluorescence. Western-blot and Real-time PCR methods were performed in normal embryonic rat group and animal models with NTDs group to detect the expression of nNOS and iNOS. Result No obvious difference was found about the expressions of nNOS in cerebral cortex of embryonic rat in the ETU group with normal brain and myeloschisis group compared with the control group(P〉0.05); The expression of nNOS in cerebral cortex of embryonic rat in the encephalocoele group decreased significantly compared with the control group(P〈0.01). iNOS expression was not detected in all groups. Conclusion nNOS might be involved in the abnormal developmental process of encephalocoele .
