摘要
β-分泌酶(β-site APP cleaving enzyme,BACE)在β位点裂解β淀粉样蛋白前体蛋白(β-amyloid precursor protein,APP)并产生β淀粉样蛋白(amyloid proteinβ,Aβ),Aβ的聚集是形成阿尔茨海默病(Alzhemier’s disease,AD)患者脑中老年斑的主要原因,引起神经元损伤和认知功能减退,在AD的发病过程中起到中心和共同通道的作用。BACE的抑制剂部分抑制BACE,可使Aβ水平下降,取得明显疗效。进一步研究BACE抑制剂的作用机制,对AD治疗具有良好的前景。
β -secretase initiates the cleavage of β -amyloid precursor protein(APP)leading to the production of amyloid- β (A β), the gathering of A β is the main cause of senile plaque in the brain of AD patient, causes neuron damage and the hypofuction of cognition, works as the center and co-gallery role in the pathogenesis of AD. The sup pressant's partial inhibition of BACE can decrease the production of A β and get manifest therapeutic effect. Further study of the way how BACE inhibitors work has favourable perspective in the treatment of AD.
