摘要
采用正交实验优化西藏变色疣柄牛肝菌粗多糖提取工艺条件,并应用气相色谱法对变色疣柄牛肝菌多糖中单糖的组成进行分析。结果表明,西藏变色疣柄牛肝菌粗多糖最佳提取条件是:提取时间3 h,温度80℃,料水比1∶15,乙醇沉淀浓度85%,此条件下多糖得率为5.008%,提取粗多糖含量为47.995%;气相色谱分析得出组成该多糖的单糖分别是葡萄糖(G lc)、半乳糖(Gal)、甘露糖(M an)、木糖(Xyl)和岩藻糖(Fuc),其摩尔比为Fuc∶Xyl∶M an∶G lc∶Gal=0.32∶0.26∶1∶14.7∶1.67。
The extraction technological conditions of polyoses of the Leccinum variicolor Wathing in Tibet were optimized by orthogonal test. The compositions of monosaccharide in polyoses which belonged to Leccinum variicolor Wathing were analysed by GC. The results showed as follows : the optimum extraction time was 3 h ;the temperature was 80 ℃ ;the ratio Of materials to water was 1 : 15 ;the deposit concentration was 85% ; the yield of polyoses was 5. 008% ,the content of extracted polyoses was 47. 995%. We found that the polyoses were composed by five kinds of monosaccharides which are Glc, Gal, Man, Xyl and Fuc by GC. The mole ratio of these monosaccharides is : Fuc : Xyl: Man: Glc : Gal = 0.32 : 0.26 : 1 : 14.7 : 1.67.
