摘要
目的观察血管抑素基因联合氟尿嘧啶腹腔化疗对抗腹腔荷瘤裸小鼠的抗恶病质效应。方法RT-PCR克隆血管抑素基因,建立重组腺病毒载体。建立人结肠癌LoVo细胞腹腔种植的模型,给予血管抑素基因联合氟尿嘧啶腹腔化疗。观察荷瘤裸小鼠的体重和摄食量的改变及血清TNF-?和IL-1水平的变化。结果血管抑素基因联合氟尿嘧啶腹腔化疗组裸小鼠的体重、摄食量、精神和活动情况明显好于对照组;治疗组的血清TNF-?和IL-1水平和肿瘤微血管密度(MVD)明显低于其他各组。结论血管抑素的基因治疗与传统的腹腔化疗联合应用,可明显的改善荷瘤裸小鼠的恶病质症状,增强其抗恶病质的效应.
Objective To study the therapeutic effect of combining angiostatin gene therapy with peritoneal chemαherapy with 5-FU on cachexia in nude mice bearing with colorectal carcinoma metastasis in abdominal cavity. Methods Angiostatin gene was cloned into adenovirus with method of RT- PCR. LoVo cell suspension was i.p to 40 nude mice. The mice were treated with combining angiostatin gene therapy and 5-FU peritoneal chemαherapy or αhers. Observation were made on the improvement of physiological conditions of the cachetic nude mice after treatment, and the serum levels of TNF-α and IL-1 were assayed by ELISA method. Result Compared with the controlled groups, the group of combining therapy was less loss of body mass, higher food intake, better mental condition and lower levels of TNF-α and IL-1. Conclusion Combining angiostatin gene therapy and 5-FU peritoneal chemαherapy has beneficial effect on cachexia in nude mice bearing with colorectal carcinoma in abdominal cavity.
