摘要
目的探讨TACO与细胞膜之间的相互作用。方法利用免疫荧光分析检测内源性TACO在细胞内的分布情况,然后利用细胞膜内膜表面展示分析和差速离心分离亚细胞组分分析检测TACO蛋白与细胞膜的结合,以及该结合与细胞膜上肌动蛋白纤维之间的相互关系。结果TACO主要分布在细胞膜和核膜附近区域。结论TACO与细胞膜具有非依赖肌动蛋白方式的结合,这一结果为阐释TACO在细胞吞噬初始阶段的功能提供了有力的实验依据。
Objective To analyse the interrelationship of tryptophan-aspartate-containing coat protein (TACO) and cellular membrane, and try to understand the molecular mechnism and function. Methods Immunofluorescence analysis was used to test the endogenous distribution of TACO in the cell. And then the inner membrane surface display analysis and differential centrifugation separation inferior celelular component was used to detect the binding between TACO and cellular membrane and their binding's relationship with the actin-binding proteins fibers. Results TACO was mainly distributed near the cellular membrane and nuclear membrane. Conclusion The binding between TACO and cell membrane is non-actin related. The result can provide evidence for exploring the function of TACO at the early stage of cell phagocytose.
