对吡喃糖氧化酶法测定血清中葡萄糖的评价 被引量:3

摘要 目的对毗喃糖氧化酶法测定血清中的葡萄糖进行评价。方法进行方法学评价试验和方法间比较试验。结果检测限为0.039mmol/L,反应进程检测低值血清498s时吸光度趋于稳定,反应吸光度为0.35A;检测高值血清10min时吸光度趋于稳定,反应吸光度为2.0A,批内CV低、中、高值分别为0.70%、0.58%、0.70%,日间CV低、高值分别为2.32%、0.63%,线性范围36.61mmol/L,平均回收率102.7%,血红蛋白14.2g/L以下、总胆红素546.65μmol/L以下、甘油三酯25.78mmol/L以下、维生素C(Vc)100mg/dL以下无显著性干扰,与己糖激酶法相关,Y=0.9906X-0.0412,r=0.9998;与葡萄糖氧化酶法相关,Y=0.9980X-0.0379,r=0.9998,相关良好。结论该法简单、稳定,与己糖激酶法和葡萄糖氧化酶法相关性良好,能较好地抗溶血、黄疸、乳糜、Vc的干扰。 To appraise pyranose oxidase method for measuring serum glucose. Methods Methodology evaluation experiment and comparison experiment between methods were carried out. Results The detective limit was 0. 039 mmol/L. In the reaction process, the absorbance tended to be stable at 498 s while measuring low value serum, and the absorbance was 0.35A. The absorbance tended to be stable at 10 min while measuring high value serum, and the absorbance was 2.0A. The CVof high, medium and low value was 0.70%, 0.58% and 0.70% for intra-run, 2.32%, 0. 63~ for inter-days respectively. The l'inear scope was 36.61 mmol/L. The average recovery rate was 102.7%. There was no significant interference when hemoglobin was lower than 14.2 g/L, total bilirubin lower than 546.65 μmol/L, triglyceride lower than 25.78 mmol/L, and vitamin C lower than 100 mg/dl. The method correlated with hexokinase (HK) assay (Y =0. 990 6X-0. 0412,r=0. 9998) and glucose oxidase(GOD)assay (Y =0. 998 0X-0. 0379,r=0. 9998). Conclusion The method is simple, stable, correlated well with HK and GOD, can resist the interference of hemolysis, jaundice, chili, vitamin C under a certain concentration.
