摘要
目的探讨LRP-5基因rs4988300位点单核苷酸多态(SNP)与多囊卵巢综合征(polycystic ovarian syndrome,PCOS)的相关性。方法采用变性高效液相色谱法检测与分析110例PCOS患者LRP-5基因rs4988300位点SNP多态性,比较肥胖与非肥胖、糖耐量受损与糖耐量正常以及血脂异常与血脂正常的PCOS患者LRP-5基因rs4988300位点多态有无差异。结果在110例PCOS患者中,共发现40例样本出现不对称峰的谱型,抽样测序,证实为LRP-5基因单核苷酸多态位点:rs4988300(G/T)。比较肥胖与非肥胖组,糖耐量受损与糖耐量正常组,血脂正常与血脂异常组之间LRP-5基因rs4988300位点多态性有无统计学差异。结论LRP-5基因rs4988300位点单核苷酸多态与PCOS糖和脂代谢异常可能无关。
[Objective] To investigate the association between single nucleotide polymorphism(SNP) of gene Lowdensity lipoprotein receptor-related protein-5 (LRP-5) at rs4988300(G/T) locus and polycystic ovarian syndrome (PCOS). [Methods] Denaturing High Performance Liquid Chromatography(DHPLC) was used to amplify the SNP of the gene LRP-5 at rs4988300(G/T) locus for all 110 PCOS patients. Comparisons were conducted in gene polymorphism between fatty group and non-fatty group, normal glucose torlerance group and inspired glucose torlerance group, and normal lipid prof'des group and altered lipid profiles group. [Results] There are 40 unsymmetric figures in all 110 samples, and the rs4988300(G/T) locus was confirmed by sampling and sequencing. There were no significant differences between fatty group and non-fatty group, normal lipid profile group and altered lipid profiles group. [Conclusion] There are no significant association between the SNP of the gene LRP-5 at rs4988300(G/T) locus and abnormal metabohsm of glucose and lipid.
