柱前衍生HPLC法测定地龙中氨基酸的含量

Determination of amino acids in Pheretima aspergillum by pre-column derivatization of HPLC

目的:分析地龙中氨基酸的含量及其特征性。方法:采用邻苯二甲醛(OPA)、9-芴甲基氯甲酸酯(FMOC)联合柱前衍生HPLC测定地龙中的17种氨基酸,峰面积外标法定量。结果:各氨基酸在26min内可得到较好分离,氨基酸浓度与峰面积呈良好的线性关系,r值均在0.995以上。结论:所建立的方法分离效果好、灵敏、准确、简便,适宜地龙药材中氨基酸的测定。

Objective:To determine the contents of the amino acids and study the characters of Pheretima aspergillum(E.Perrier). Methods:The samples of Pheretima aspergillum(E.Perrier) were derived with o-phthaldialdehyde (OPA) and 9-fluorenylmethyl chloroformate (FMOC) in pre-column , and separated by HPLC. Seventeen kinds of amino acids were determined with the ration of peak area by external standard calibration.Results:All kinds of amino acids can be well separated in 26 min,the concentration of amino acids and peak areas achieved a nice linear relations ( r is higher than 0.995).Conclusion:The method is sensitive,accurate,simple,and reliable for the determination of amino acids in Pheretima aspergillum(E.Perrier).
