摘要
目的:观察雷帕霉素(rapamycin)对培养的人Tenon′s囊成纤维细胞的增殖及细胞周期的作用。方法:将培养的人Tenon′s囊成纤维细胞用含不同雷帕霉素浓度的培养液培养24 h后,用MTT法测定细胞的增殖情况,用流式细胞仪测定细胞周期的变化情况。结果:用含10、20、40、80和160 ng/ml雷帕霉素的培养液培养细胞时,细胞增殖抑制率分别为9.32%,22.55%,41.42%,42.25%,43.57%。细胞周期分析显示,雷帕霉素(40 ng/ml)作用后,G0/G1期细胞较对照组增多(41.76%vs 44.19%,P<0.05),S期细胞较对照组减少(45.42%vs 40.18%,P<0.05)。结论:雷帕霉素具有抑制人Tenon′s囊成纤维细胞增殖的效果,且随浓度增加抑制作用增强,并将细胞抑制在G1期的限制点内。
Objective: To explore the effect of rapamycin on the proliferation and differentiation of human Tenon's capsule fibroblasts in cell cycles. Methods: The cultured fibroblasts were treated in DMEM medium containing various concentrations of rapamyein for 24 hours. The growth of fibroblasts was measured by a methylthiazlyl tetrazolium (MTT) assay. The flow cytometry method was used to evaluate the cell cycles. Results: Proliferation of fibroblasts were suppressed by rapamycin from a dose of 10 ng/ml to 160 ng/ml( P 〈 0.05). The cell growth inhibition rate treated with mpamycin concentrations of 10 ng/ml, 20 ng/ml, 40 ng/ml, 80 ng/ml, and 160 ng/ml were 9.32%, 22.55%, 41.42%, 42.25%, and 43.57% respectively. The cell population was significantly increased at the G0/C1 phase from 41.76% to 44.19% and decreased at the S phase from 45.42% to 40.18% after the rapamycin treatment. Conclusion: Rapamycin is a potential inhibitor that blocks the proliferation of human Tenon's capsule fibroblasts at the C1 restriction point.
