幽门螺杆菌外膜蛋白基因omp22克隆测序及生物信息学分析 被引量:3

摘要 [目的]克隆幽门螺杆菌(helicobacter pylori,Hp)外膜蛋白omp22基因,并对其进行测序及生物信息学分析,为幽门螺杆菌疫苗研究提供依据。[方法]利用PCR技术从Helicobacter pylori郑州分离株MEL-Hp27染色体DNA上扩增出omp22基因,将其定向插入克隆载体pBluescriptbⅡ,并进行序列测定和生物信息学软件分析其生物学特性。[结果]用PCR方法扩增的omp22基因长度为540bp,编码179个氨基酸,DNAstar和Omiga软件预测其蛋白质的相对分子量(Mr)约为20kDa,并显示出良好的抗原性和疏水性。[结论]本研究获得了序列正确的幽门螺杆菌omp22基因,为其重组蛋白表达及其相关研究奠定了基础。 [Objective] TO clone, sequence the omp22 gene of Helicobacter pylori and analyze the biological information, to provide scientific basis for research work Of Hp vaccine. [Methods] omp22 gene of the Helieobacter pylori MEL-Hp27 was amplified by PCR. After being purified, the target fragment was cloned into plasmid pBluescriptb Ⅱ and was sequenced, The biological property at the amino acid level was analyzed by DNAstar5.0 and Omiga2.0. [Results] omp22 gene of 540 bp, coding the polypeptides of 179 amino acids, was obtained fi'om the Helicobaeter pylori strains MEL-HP27 genome DNA .Tile bi- ological information software predicted its relative molecular mass (Mr) was about 20kDa and possessed good antigeneity and hydrophobicity. [Conclusions] A eonfimled omp22 gene has been cloned, which providing a good foundation for recombinadon, expression and related study.
