宫颈鳞癌中P120ctn及相关蛋白的表达及临床意义

摘要 目的:研究P120连环蛋白(P120ctn)、β连环蛋白(β—cat)、E钙粘蛋白(E—cad)及Ki67蛋白在宫颈鳞癌中的表达及临床意义,分析P120ctn异常表达与B—cat、E—cad及Ki67蛋白异常表达的相互联系。方法:应用免疫组化SP法检测P120ctn、β—cat、E—cad及Ki67蛋白在26例正常宫颈组织和52例宫颈鳞癌组织中的表达。结果:①正常宫颈组织中P120ctn、β—cat及E—cad表达在胞膜,胞浆少见;宫颈鳞癌中P120ctn及E—cad胞膜表达降低,出现胞浆表达,异常表达率分别为41/52(78.8%),38/52(73.0%);β—cat除了胞浆表达以外,还伴随胞核表达,异常表达率为39/52(75%);P120ctn、β—cat及E—cad异常表达与肿瘤细胞分化程度、淋巴结转移有关(P〈0.05),与年龄、临床分期无关(P〉0.05);②同一组织标本中P120ctn表达异常,同时伴β-cat及E—cad异常表达;③正常宫颈组织中Ki67表达率为4/26(16%),宫颈鳞癌组织中表达率为40/52(80%);Ki67异常表达与临床分期有关(P〈0.05),与年龄、肿瘤细胞分化程度及淋巴结转移无关(P〉0.05);P120ctn异常表达与Ki67蛋白异常表达有相关性。结论:P120cm、β-eat及E—cad在宫颈鳞癌中异常表达,三者异常表达有很好一致性,P120ctn异常表达与Ki67蛋白异常表达有相关性。P120ctn及相关蛋白异常表达可作为宫颈鳞癌诊断的辅助指标,并为判断疾病发展及转移提供客观依据。 Objective: To research the relationship between abnormal expression of P120 - catenin and clinic pathologic significance in squamous cervical carcinoma(SCC) , β - catenin, E - cadherin and Ki67 protein. Methods: Expression of P120 - catenin, 13 - catenin, E - eadherin and Ki67 protein were examined in 26 cases of normal cervical epithelia samples and 52 cases of SCC by immunohistochemical staining (SP method). Results: ① In the normal cervical epithelia tissue, P120 - catenin, β - catenin and E - cadherin expression showed on the membrane and lowly in the cytoplasm; In the SCC, P120 -catenin and E - cadherin expression were found in cytoplasm, lower in the membrane, abnormal exoression rates were 41/52 (78. 8% ), 38/52 (73. 0% ) respectcticely; β -catenin expression showed in the cytoplasm and nuclea, abnormal expression rates was 39/52(75% ) ; Abnormal expression of P120 - catenin, β - catenin and E - cadherin were significantly correlated with status of tumor differentiation and lymph node metastasis(P 〈 0.05 ) , no significant association was found between abnormal expression and clinical stage and age( P 〉 0.05 ). ②P120 - catenin abnory expressed, as well as β - catenin and E - cadherin in identical organization imen. ③ In normal cervical tissue, expression rates of Ki67 protein was 4/26 ( 16% ) , In SCC , the expression rates was 40/52 (80%) ; Abnormal expression of Ki67 protein was significantly correlated with clinical stage, no significant correlated with tumor differentiation status, lymph node metastasis and age (P 〉 0.05 ) ; There are relationship between abnormal expression of P120 - catenin and Ki67 protein. Conclusions: In SCC, P120 - catenin, β - catenin and E - cadherin showed abnormal expression, the expressions of three proteins above have highly conformance, there are relationship between abnormal expression of P120 -catenin and Ki67 protein, abnormal expression of P120ctn and related proteins may be useful markers for diagnosis of SCC, and provide a objective fact for assessing development and metastasis of scc.
